Genetic informationFloxed mouse line for interleukin 6 (Il6). To this end, exon 2 from Il6 gene has been flanked with loxP sites.
Phenotypic informationWith this number of backcrosses (ca. 5) the floxed mice show some phenotype, since the floxed Il6 gene is expressed less than in normal wild-type mice upon endotoxin administration. We have already produced some conditional KO mice with several cre-expressing mouse lines and there is a clear difference between floxed and KO mice regarding Il6 levels.
Breeding historyNeo cassette was removed by crossing with flp-expressing mice once one of the chimeras was found to have germ line transmission upon crossing with C57BL/6. The flp gene was removed upon crossing with C57BL/6. Finally, offspring mice positive for the floxed gene were crossed with each other and a homozygous line of these floxed mice is maintained that way in the general animal services of this university. We have started to further backcross this line with C57BL/6 in the Animal Physiology unit; in total we are about in the fifth generation.
