Tsien/San Diego) in opposite orientation to Gt(ROSA)26Sor transcription and downstream of a loxP-flanked stop sequence (promoterless neomycin resistance gene and three copies of the bovine growth hormone polyadenylation sequence). This entire construct was inserted into the Gt(ROSA)26Sor locus via gene targeting of C57BL/6-derived Bruce 4 embryonic stem cells. |
Phenotypic information | Mice that are homozygous for the mutant allele are viable, fertile, normal in size and do not display any gross physical or behavioral abnormalities. Homozygous females exhibit a somewhat reduced fertility when compared with C57BL/6 wildtype animals. The mice contain a tandem-dimer red fluorescent protein (tdRFP) gene inserted in opposite transcriptional orientation into the Gt(ROSA)26Sor locus. Expression of tdRFP is prevented by an upstream loxP-flanked STOP sequence. When bred to mice expressing a cre recombinase gene under the control of a promoter of interest, the tdRFP cassette is irreversibly flipped and the STOP sequence deleted, resulting in tdRFP expression in the cells/tissues of interest. These mutant mice may be useful in monitoring cre recombinase expression in living tissues cytofluorimetrically and histologically. They have been generated in particular for multicolor lineage tracing studies in combination with GFP-based fluorescent proteins. |
Breeding history | The mutation was generated by targeting the appropriate construct into the Gt(ROSA)26Sor locus of C57BL/6-derived Bruce4 embryonic stem cells. Correctly targeted ES cells were microinjected in BALB/c blastocysts. One chimeric animal was bred to a C57BL/6 mouse. Mutant progeny were intercrossed to generate homozygous mutants. The strain is maintained by intercrossing homozygous mutants. |
