Genetic informationTo generate Miwi2 null allele we recombineered a targeting construct in which exon 17 was flanked with loxP sites. Cre-mediated deletion of exon 17 results in out of frame splicing between exon 16 and 18 resulting in stop codons prior to the last exon, targeting the mutant transcript for nonsense mediated decay (NMD). Should a fraction of the mutant transcript escape NMD, a truncated protein would be made that lacks most of the Piwi domain and thus would likely be non-functional.
Phenotypic informationHomozygous male mice for Miwi2 null allele are viable but present impairment of spermatogenesis that results in severe infertility. The mutation does not affect female fertility at all.
Breeding historyMice carrying the targeted allele were crossed with Cre expressing transgenic mice (CreeR) to remove the LoxP-flanked NeoR cassette. The resulting offspring was backcrossed to C57BL/6N to remove the Cre.
