Genetic informationThe transgenic construct expresses human NR0B2 (former SHP) (HA-Tag) flagged under the mouse Ttr promoter. Hepatic SHP expression in this transgenic line is 7-fold above endogenous levels.
Phenotypic informationHomozygous:
The livers of transgenic animals are visibly enlarged and pale in color. Extensive lipid accumulation is detected in SHP-expressing hepatocytes. Hepatic triglyceride levels are markedly increased and the concentration of hepatic bile acids is significantly decreased. Major defects in Tg-SHP mice occur in bile acid synthesis, lipogenesis, de novo cholesterol biosynthesis and absorption pathways.
