Genetic information | Conditional knock-in of the Tid (Dnaja3) SC splice form (GenBank Acc. Nr.: AF326358; PubMed ID:11116152) into Gt(ROSA)26Sor, homozygous. The cDNA encoding the Tid splice form was cloned into the pRAPIDflirt vector. The structure of the targeting vector is as follows: Gt(ROSA)26Sor::CAG–NeoR–WSS/STOP–Tid splice form-IRES–EGFP. The NeoR-WSS/Stop cassette is floxed. Thus, the activation of Tid expression only occurs after cre-mediated excision of the cassette. Because of the EGFP-coding sequence the mutant mice express it, too. To generate the knock-in mutant line Bruce 4 ES cells (C57BL/6-derived) were used. Recombinant ES cells were injected into C57BL/6 albino blastocysts. For GLT chimeric males were mated with C57BL/6 albino females. |
