Genetic informationTransgenic mouse strain expressing mouse Hgf under the Wap promoter.
Phenotypic informationWAP-HGF transgenic females developed hyperplastic ductal mammary trees and multifocal invasive mammary tumors after several pregnancies, some of which progressed to lung metastases. The WAP gene promoter, although active during estrus, is prominently induced during pregnancy and remains active throughout lactation in mammary glands. Consistent with this, continuous expression of HGF through successive pregnancies resulted in the appearance of mammary adenocarcinomas with areas of squamous metaplasia. WAP-HGF tumors expressed HGF and displayed c-Met phosphorylation. Histological and immunohistochemical analyses of sections confirmed that c-myc and Akt were activated in WAP-HGF tumors. WAP-HGF tumors presented malignancy markers like osteopontin expression and were progesterone receptor-negative. Stabilization of beta-catenin was found in areas of squamous metaplasia.
Breeding historyThe strain was kept in an FBV genetic background for 10 generations. Sperm was frozen and cryo-preserved for 6 years, the strain was then regenerated by IVF with wild-type FVB females.
