Genetic informationChromosome mediated gene transfer was used to insert human chromosome 21 from a human cell line into mouse ES cells and hence into mice.
Phenotypic informationAffected mice suffer from learning abnormalities, reduced long-term potentiation in the hippocampus, heart and brain developmental defects, craniofacial dysmorphology (this strain models human Down syndrome).
Breeding historyThe extra human chromosome 21 is only transmitted through the female germline, so the strain is maintained by crossing Tc1 females to male B6129S8F1 mice. Transmitting females only transmit the human chromosome to up to 40% of their offspring on this genetic background.
