Genetic informationExons 2 and 3 flanked with loxP and ES clones transfected with cre recombinase. ES clones with deleted exons 2 and 3 where injected to produce the RhoC knock-out. Deletion of RhoC exons 2 and 3 should introduce termination codons into all three reading frames and remove 70% of the protein sequence.
Phenotypic informationRhoC+/- mice were then intercrossed to obtain homozygous RhoCDelta2-3/Delta2-3 (RhoC-/-) mice. RhoC-/- mice were born at the expected Mendelian ratio and were viable and fertile. RhoC is essential for metastasis but not for primary tumor formation.
