Genetic informationTo generate the Gt(ROSA)26Sor-mCherry-Rpl10a allele, we PCR amplified mCherry from mCherry-pRSET-B (provided by Roger Tsien), and Rpl10a from CS-EGFP-L10A (provided by Nathaniel Heintz). mCherry was digested with NheI/XhoI, Rpl10a with XhoI/NotI, and subcloned into the NheI/NotI sites of pBSApBpACAGftILn. Finally, the conditional expression construct was released with PacI and AscI, and subcloned into the PacI and AscI sites of pRosa26PAS. The construct was linearized and electroporated into F1 ES cells. Colonies were screened by PCR and positive colonies expanded and confirmed by PCR. One targeted clone was injected into host C57BL/6J (The Jackson Laboratory) blastocysts by the Karolinska Center for Transgene Technologies, Karolinska Institutet.
