Genetic informationThe cre-inducible tdRFP cassette was targeted into the Gt(ROSA)26Sor locus of Bruce4 ES cells, which are derived from mice of the widely used inbred C57BL/6 strain. Insertion of a codon-improved cre (icre) recombinase into a bacterial artificial chromosome carrying the Ltf gene (BAC; RP24-166N8; purchased from Children's Hospital Oakland Research Institute, Oakland, CA) was performed via homologous recombination in E. coli. A cassette containing the icre recombinase, an artificial intron, a bovine growth hormone polyadenylation signal and an ampicillin-resistance gene flanked by Frt sites, was recombined into the first exon of the Ltf gene. To delete the ampicillin gene, a plasmid expressing the FLP recombinase was transformed into E.coli harboring the recombined BAC. Correct insertion and ampR-gene deletion were verified by Southern blotting and by DNA sequencing. BAC was purified using a CsCl-gradient and ultracentrifugation and injected into the pro-nuclei of (B6 x 129/Sv)F1 oocytes.
Phenotypic informationHomozygous:
RFP mice show red fluorescence; Ltf-cre: no homozygous mice produced.

Heterozygous:
No overt phenotypes in gross morphology, life expectancy or breeding efficiency, nor did we observe any organ pathologies during a breeding period of >36 months
Breeding historyAnimals with suited genotype were selected, independent if they were sibs or not.
