Genetic informationHomologous recombination of the floxed Tid (Dnaja3) allele, homozygous. To generate the conditional Tid (Dnaja3) knock-out line a targeting vector harbouring the mouse Tid genomic region, with a NeoR-cassette and FRT and loxP sites was generated. The genomic region was cloned into the pCR4BluntTopo vector. Exons 2 and 3 have been floxed, as a NeoR-cassette equipped with a loxP-FRT site (5’) and a FRT site (3’) has been introduced in intron 1 and a further loxP site has been located in intron 3. To generate the original Tid-flox line TC-1 (129S6/SvEvTac) ES cells (PolyGene AG, Riedmattstrasse 9, 8153 Rümlang, Schweiz) were used. Recombinant ES cells were injected into C57BL/6 albino blastocysts. For GLT chimeric males were mated with C57BL/6 albino females. Mutant offspring of this cross was further propagated by inbreeding. The conditional Tid knock-out line (line 4) was generated by flipping out of the NeoR-cassette, upon crossing it with Flpe deleter mice (B6;SJL-Tg(ACTFLPe)9205Dym/J strain). Line 5 originates from line 4, by backcrossing to the C57BL/6 background.
