| Genetic information | This is a knockout mouse line with a targeted inactivation of Pcbp4 gene. This mouse gene is located on chromosome 9 and extends on around 10 kb. The gene is composed of 13 exons. The cDNA contains 1984 nucleotides. The open reading frame encodes for a 403 amino acid polypeptide. The translation initiation site is located in exon 2 and the stop codon in exon 13. The constitutive knockout line was generated using a conditional gene targeting strategy (“Safe knock-out” approach from GenOway). Using homologous recombination, the targeted region (exons 2 to 12 containing the 3 KH functional domains of the PCBP4 protein) was flanked by two loxP sites. Targeted GenOway 129/SvPas ES clones were generated and injected into C57BL/6 blastocysts. Chimeras were crossed with C57BL/6 CMV-cre mice to induce a heterozygous deletion of Pcbp4. The animals were intercrossed to generated homozygous Pcbp4 KO mice. |
