Genetic informationLgals1 (Gal-1; Lect14) knock-out mice were obtained by kind gift of Dr. F. Poirier (Poirier F. and Robertson E. Development 119, 1229-1236; 1993). They were generated by targeted mutation. In particular, exon 2, encoding the carbohydrate-binding domain, of Lgals1 locus was replaced with a neomycin selection cassette. Western blot analysis and immunostaining of sections showed an absence of encoded protein in muscle tissue obtained from adult homozygous mutant mice.
Phenotypic informationHomozygous:
We did not detect any abnormalities in the breeding or viability of these mice. Alteration of the mouse olfactory system has been reported in other genetic backgrounds (129S/SvEv * MF1; Puche AC. et al., Dev Biol 1996). In particular, while olfactory nerve itself was normal, projections from it to the dorsocaudal surface of the olfactory bulb were severely reduced and axons never terminated in glomeruli, but we have not analysed the phenotype at this level in our C57BL/6J genetic background.

Breeding historyOriginal background of the Gal-1 -/- mice was 129Sv (original background strain from Françoise Poirier). Dr. Poirier later generated them into a C57BL/6J background by backcrossing to C57BL/6J wild-type mice. We have maintained these mice into this genetic background by inbred crossing (B6 Gal-1 -/- x B6 Gal-1 -/-) as well as periodic breeding with C57BL/6J wild-type (once a year, to avoid excessive inbreeding).
