Genetic informationTatdn1 loxP/loxP was designed and produced by ARMI/MONASH, Australia. Tatdn1 exon 3 was flanked by loxP sites. Deletion of exon 3 induces a frameshift and stop codon in exon 4, giving a protein of 40 amino acids. The neomycin resistance cassette for ES selection was originally flanked by FRT sites (FRT-PGK-em7-Neomycin-pA-FRT). Male chimeras were crossed with C57BL/6 females in 2011. Later, we crossed Tatdn1 transgenic mice with a Gt(ROSA)26Sor-FLPe strain, inducing deletion of the NeoR cassette and the present Tatdn1 loxP/loxP mice lack the NeoR gene.
Phenotypic informationHomozygous:
Fertility and embryonic development seem not affected. Deletion of Tatdn1 by further crossing with cre recombinase-expressing mice produces normal fertile mice, breeding at the expected number of male and female descendants. Normal gross morphology. We have analysed heart function and behaviour. Mild ventricular hypertrophy and potential locomotor alterations are induced by Tatdn1 deletion.

Breeding historyTatdn1 loxP FRT male chimeras were crossed with C57BL/6 wild type females to obtain the F1 mice in 2011. Floxed mice were crossed for several generations within the C57BL/6 background. FRT deletion was generated by crossing with the Gt(ROSA)26Sor-FLPe strain from the Jackson Lab. (B6.129S4-Gt(ROSA)26Sortm1(FLP1)Dym/RainJ) in 2013. Since then, animals have been backcrossed in a C57BL/6 background many times.
