| Genetic information | The 9.0 kb promoter of tyrosine hydroxylase (pTH) gene was used to direct the expression of rat glucose-6-phosphate dehydrogenase (G6PD) into catecholaminergic regions. Rat G6PD coding sequence was cloned by RT-PCR using 2FG6PD as forward primer: 5'-TGGCAGCGGCAACTAAAT-3' and 1654R as reverse primer: 5'-TAGAGGGTTAGATGGTGA-3'. A 11.5 kb HindIII/NotI fragment, containing the pTH plus the G6PD coding sequence flanked by the beta-globin intron II and a poly A signal was cut from pcDNA3-TH/G6PD plasmid and used for injection into fertilized C57BL/6 eggs to generate pTH-G6PD transgenic founder animals. Multiple copies of G6pd were inserted in the pTH-G6PD (5BB) mouse genome. |
