Genetic informationThe following elements were inserted into intron 2 to create this knockout-first allele: an FRT site, the En2 intron, sequence coding for 2A self-cleaving peptide, mTagBFP2 reporter gene coding sequence, SV40 poly(A) signal sequence, a loxP site, a neomycin resistance gene cassette, a second FRT site and a second loxP site. A third loxP site was inserted into intron 3. Exon 3 (ENSMUSE00000161960) is thus flanked by loxP sites. A "conditional-ready" (floxed) allele can be created by flp recombinase expression in mice carrying this allele. Subsequent cre expression results in a knock-out mouse. If cre expression occurs without flp expression, a reporter knock-out mouse will be created.
