Genetic informationA loxP site was inserted upstream exon 22 (ENSMUSE00000457495). A lox511 site, a 531 bp synthetic DNA sequence consisting of a degenerated and inverted version of exon 22 harboring the mutation of interest flanked by human intronic sequence, a reverse loxP site, a FRT-flanked neomycin resistance cassette and a reverse lox511 site were inserted downstream exon 22. The neo cassette was removed through subsequent flp-mediated recombination leading to a conditional PM allele. Subsequent cre-mediated recombination will change asparagine codon 858 (AAC) to serine (TCT) (C.2572_2574delAACinsTCT p.N858S).
