| Genetic information | A PGK-HPRT minigene replaced 414 bp of the promoter region and exons 1 and 2 of the keratin 10 (Krt10) gene via homologous recombination in ES cells. |
| Phenotypic information | Krt10 -/- mice were born at the expected Mendelian ratio, were fully viable, and showed no obvious skin defect resulting from birth stress. Histological analysis of neonatal skin revealed normal stratification and an unaltered number of epidermal cell layers, although a slight increase in granular cell size and the size of keratohyalin granules and a loose appearance of the stratum corneum was observed. No cytolysis in Krt10 -/- epidermis was seen, suggesting that filaments consisting of Krt10 and 1 are not essential for maintaining the integrity of suprabasal epidermis in these mice. Thus, most probably a compensation by basal keratins 5, 14, and 15 prevents cytolysis and permits normal epidermal differentiation in Krt10 -/- mice. |
