Genetic informationThe construct is a CMV-beta plasmid, with the beta-galactosidase replaced by HA-tagged murine MK5. We mutated this MK5 from Leu337 to Ala. This mutation renders the kinase constitutively active. Random integration.
Phenotypic informationNo major defects, a very low amount of mice had tumours (2 founder mice), some mice had microbial infections (especially line A was prone). Whilst background breeding, mutated mice seemed more anxious compared to WT, a feature that disappeared the longer the background breeding lasted.
Breeding historyThe linearized construct was microinjected into CBA x C57BL/6 fertilized eggs. The resulting mice positive for the transgene bred to wt C57BL/6 for 6 generations.
