| Genetic information | To generate the transgenic mouse lines, we used constructs containing 254 bp of the human HKII promoter driving 42% of the maximal basal expression of the firefly (P. pyralis) luciferase reporter cDNA in vitro, as described previously by Malkki et al. (1997). For microinjection, DNA fragments encompassing 254 bp of the HKII promoter, 106 bp of HKII exon I noncoding sequence, the luciferase coding sequence (1.8 kb), and small T intron and polyadenylation sequences of SV40 (1.4 kb) were released by digestion with PstI. |
