Genetic informationThis mutation carries a 5 bp insertion (ATCAG) after nucleotide 1791 of mRNA, resulting in frame-shift in the Ptprc coding sequence, introducing 26 new amino acids and premature termination.
Phenotypic informationThe loc/loc mice have decreased expression of CD45 on CD19+ B cells but normal numbers of B cells. The strain also exhibits slightly reduced levels of IgM+IgD-, IgM+IgD+ and IgD+CD21+ cells. Reduced levels of B220loCD19+ cells but normal levels of B220+CD19+ in bone marrow. A 3-fold decrease in CD4+CD8+, double-positive (DP) thymocytes and a 10-fold decrease in CD4+ single-positive (SP) thymocytes. A 5-fold decrease in CD8 SP thymocytes. DP thymocytes had a 2.5-fold increase in expression of CD4 and CD8. Cell intrinsic co-receptor expression on SP T cells was increased 1.5-fold. There is a dramatic decrease in CD8+CD44lo/med cells. TCRa is up-regulated on DP thymocytes and down-regulated on mature T cells. CD5 is down-regulated on DP thymocytes and up-regulated on peripheral T cells. The decrease in DP thymocytes is not cell intrinsic but the decrease in SP thymocytes is cell intrinsic.
