| Genetic information | To generate the transgenic mouse lines, we used construct containing 4077 bp of the human HKII promoter, driving 100% of the maximal basal expression of the firefly (P. pyralis) luciferase reporter cDNA in vitro, as described previously by Malkki et al. (1997). For microinjection, DNA fragments encompassing 4077 bp of the HKII promoter, 106 bp of HKII exon I noncoding sequence, the luciferase coding sequence (1.8 kb), and small T intron and polyadenylation sequences of SV40 (1.4 kb) were released with XhoI after introduction of the XhoI restriction site to replace the upstream PstI site by recloning and ligation of the oligonucleotide linker. The 4077 bp construct contained an internal PstI site. |
