| Phenotypic information | The 5-HT2B-/- mice were reported (Nebigil et al, 2000) to display midgestation and postnatal partial lethality due to cardiac defects. The 5-HT2B receptor-mutant mice that survived the first postnatal week (about 50%) developed to adulthood with cardiac problems, though these become compensated with age (Jaffre et al, 2009) and 10-20 weeks old mice have no more detectable defects. The 5-HT2B receptor-mutant mice display no basal pulmonary phenotype, instead they are resistant to inducers of pulmonary hypertension (Launay et al, 2002). So the cardiovascular and pulmonary phenotypes are undetectable in living adult mice. |
| Breeding history | The targeting vector was constructed with the selective bacterial neo gene introduced into the exon 2 sequence of the 5-HT2B receptor locus. This resulted in a non-functional allele of 5-HT2B as it interrupts the protein reading frame. One recombination event was obtained after screening of about 4000 ES cell clones. Southern blot analysis on the recombined clone using a probe derived from neo sequence revealed that it contained a single expected 16 Kbp fragment in an HpaI restriction enzyme digestion excluding multiple insertion of the targeting vector. From this recombined 5-HT2B ES cell strain, 13 chimeric animals were created by injection into C57BL/6 blastocyst embryos. These chimeras were bred to either C57BL/6NCrl or 129S2/SvPas mouse strain to obtain germ-line transmission. |
