Genetic informationA PGK-neo cassette replaced the first coding exon, which contains the ATG as well as the PLSTIRE domain essential for cyclin binding. PCR confirmed recombination in mutant mice. Immunohistochemical analysis of lymphocytes indicated lack of protein expression in mutants.
Phenotypic informationHeterozygous animals do not exhibit any phenotype. Homozygous mice show abnormal hematopoietic system morphology/development, decreased T cell proliferation, abnormal spleen and thymus morphology and reduced female fertility (approximately 30% of the females are sterile).
Breeding historyChimeras were initially crossed with CD1 females for germ line transmission analysis. The line has been backcrossed into C57BL/6J for 10 generations.
