Genetic informationThe strain was obtained by microinjection of the genetrap (GGTC) D032B11 clone in C57BL/6J blastocyts. In this clone the genetrap vector inserted in intron 2, 1.52 kbp upstream of exon 3. The gene trap vector has pairs of FRT, F3, loxP and lox5171 sites allowing for inversion of the trapping cassette. Before Cre or Flp mediated recombination the allele is functionally equivalent to a knockout allele.
Phenotypic informationNo phenotype in heterozygosis. Crosses between heterozygous animals failed to produce live Cdk7loxfrt/loxfrt offspring as well as E13.5 and E9.5 embryos. Homozygous embryos die at peri-implantation stage. Analysis of E5.5 embryos derived from these crosses revealed deciduas with a high percentage of apoptotic cells within the embryonic body that were negative for Cdk7 immunostaining.
Breeding historyChimeras were crossed with C57BL/6J females for germ line transmission analysis. The line has been maintained by crossing heterozygous animals with C57BL/6J mice or by crossing heterozygous animals among themselves.
