| Genetic information | The strain was obtained by microinjection of the genetrap (GGTC) D032B11 clone in C57BL/6J blastocyts. In this clone the genetrap vector inserted in intron 2, 1.52 kbp upstream of exon 3. The genetrap vector has pairs of FRT, F3, loxP and lox5171 sites allowing for inversion of the trapping cassette. In this strain, Flp mediated recombination inverted the genetrap vector in intron 2 allowing expression of the gene from the Cdk7lox allele. Upon Cre-mediated recombination, the genetrap vector can be inverted again interfering with gene expression. The Cdk7lox behaves like a Cre/lox conditional KO allele in this strain. |
| Phenotypic information | No phenotype in heterozygosis or homozygosis. The allele behaves as a true conditional knockout allele for Cdk7. |
| Breeding history | Chimeras were crossed with C57BL/6J females for germ line transmission analysis. Heterozygous animals carrying the original genetrap allele were crossed with a transgenic line expressing the Flp recombinase in the germ line (Rodríguez et al., 2000; PMID:10835623). The line has been maintained by crossing homozygous animals or heterozygous animals among themselves. |
