Genetic informationThe insertion of a neomycin resistance cassette disrupted the codon for the third amino acid of the mature protein, located in the third exon. Immunoblotting and immunohistochemistry were both unable to detect protein in mutants.
Phenotypic informationNervous system: reduced long term potentiation (PubMed ID: 8587606). Strong inhibition of LTP in the dentate gyrus but not in CA1, however spatial learning as assessed in the water-maze is unimpaired. Respiratory system: pulmonary fibrosis (PubMed ID: 16049324). Marked increase in severity of lung fibrosis and more extensive collagen deposition in response to intra-tracheal bleomycin, compared to controls.
Breeding historyGene targeted in 129/SvEv stem cells, offspring outbred to C57BL/6J mice, then backcrossed for 4 generations onto 129/SvEv before being maintained by interbreeding.
