| Genetic information | The mouse Mmp28 gene was cloned from the 129/Sv genome (gene ID: 118453). To construct the homologous extensions of the targeting construct, a 4.8-kb EcoRI-XbaI fragment containing the 3 prime terminus of intron 1, exons 2 and 3, and 4 kb of intron 3 and a 2.1-kb XhoI-EcoRI fragment containing the 3 prime untranslated end of exon 8 and downstream sequence was isolated. A pgk-neomycin resistance cassette was inserted between the two arms and a diphtheria toxin (dta) cassette was ligated to the 3 prime end. The targeting construct was linearized with XhoI and introduced into RW-4 ES cells. |
| Phenotypic information | Homozygous:Homozygous MMP28-deficient mice have no overt phenotype. However, using a murine model of Pseudomonas aeruginosa pneumonia, it was found that Mmp28-/- mice had an early increase in macrophage recruitment into the lungs, as well as enhanced bacterial clearance and reduced pulmonary neutrophilia due to accelerated macrophage influx.Heterozygous:Heterozygous mice do not show an obvious phenotype. |
| Breeding history | 10 backcrosses to C57BL/6N. |
