| Genetic information | This transgenic line expresses a mouse Tnf gene containing a deletion mutation of the codons encoding the 1st to the 12th amino acids of the mature 17-kDa TNF protein. This mutant TNF gene has previously been shown to produce a transmembrane TNF protein that remains bioactive against cellular targets in vitro and in vivo. Approximately 50 copies of the transgene were inserted. Northern analysis demonstrated transgene expression in thymus, mesenteric lymph nodes, spleen, kidney, liver, heart, brain, skin and joints. The expressed protein was undetectable in serum as determined by ELISA assays. |
| Phenotypic information | Homozygous:N/AHeterozygous:The mice develop chronic inflammatory arthritis, with 100% phenotypic penetrance, starting at 3-4 weeks of age. The arthritic phenotype is TNFRI-dependent and it is significantly delayed in the absence of the p75TNFR. |
