Genetic informationThe AirnT allele contains a 1.2kb fragment from the rabbit beta-globin gene (bp 31392-32590; M18818) inserted 3kb downstream from the Airn TSS that causes the Airn long non-coding (lnc) RNA to be truncated and lose its imprinted silencing function. A 9.6kb BglII fragment from cosmid 940PS (129/Sv) containing the Airn promoter (mm10 chr17:12,740,025-12,749,644) was used to make the targeting construct. Targeting was done in E14 ES cells (129/Ola) and ES cells were injected into C57BL/6 blastocysts to generate chimeric mice.
Phenotypic informationHomozygous:
The phenotype of homozygous mice is the same as for heteroyzgous mice with paternal inheritance, as Airn is only expressed from the paternal allele.

Heterozygous:
Heterozygotes with a paternally inherited AirnT allele show a 15% reduction in birth weight due to biallelic expression of Igf2r, as truncated Airn can no longer silence the paternal copy. Maternal heterozygotes show no phenotype, as Airn is not expressed from the maternal allele.
Breeding historyAirnT mice were derived from 129/Sv x C57BL/6 chimeras and then backcrossed to FVB/N for over 50 generations.
