Genetic informationMutagenesis of seven Otx2 binding sites (OBS) on Sox2 promoter in the region spanning to -4033 to -2408 respect to methionine 1. The mutations are generated on the basis of chip-seq data that identify a region of Sox2 promoter spanning -4033 to -2408, respect to methionine 1, containing seven putative binding sites of Otx2 protein. These binding sites were abolished by PCR mutagenesis, the region cloned into targeting vector and introduced in ES via homologous recombination.
