Information from provider

ProviderMineko Terao
Provider affiliationLaboratory of Molecular Biology, Department of Biochemistry and Molecular Pharmacology, Istituto di Ricerche Farmacologiche "Mario Negri"
Genetic informationInsertion of transgene expressing mouse mutated xanthine dehydrogenase cDNA (R883M) on chromosome 1qA3.
Phenotypic informationHomozygous:
The size and the body weight of the homozygous animals are significantly reduced relative to the wild-type and heterozygous animals. After weaning, ocular abnormalities, such as diffused corneal opacity, increased volume of the eye globe (buphthalmos), corneal edema and elevation of intraocular pressure (IOP), become progressively evident. Histologically, the eyes of homozygous animals show the complete closure of the filtration angle, corneal lesions and neuroretinal atrophy. Dysgenesis of the anterior segment and filtration angle closure lead to glaucoma.

Heterozygous:
The phenotype of heterozygous animals is similar to that of the wild-type animals, except for IOP, which is slightly higher than that of wild-type counterpart.
Breeding historyThe transgenic line was originally created in BDF1 and backcrossed for at least 10 generations. The background was subsequently changed by mating with C57BL/6 for at least 15 generations. The substrain used for backcrossing of our glaucomatous mice was C57BL/6JOlaHsd (Envigo).
