Genetic informationEmbryonic stem (ES) cells transfected with a Slc20a2 targeting vector were purchased from the International Knockout Mouse Consortium (IKMC)/ European Conditional Mouse Mutagenesis Program (EUCOMM). In these ES cells, the exon 4 of the sodium/phosphate cotransporter Slc20a2/Pit-2 was flanked by loxP sites. Upstream of the 5’ loxP site, the vector contained a cassette for production of beta-galactosidase, followed by a neomycin phosphotransferase gene cassette. ES cells were sent to the Institute of Labortierkunde (LTK) of the University of Zurich for morula aggregation and implantation into foster mothers. Upon breeding to homozygosity, floxed/floxed mice were crossed with flp recombinase full deleters (64FlpB6) for removal of the beta-gal/neo cassettes.
Phenotypic informationHomozygous:
An intestinal-specific knockout was generated by breeding floxed Slc20a2 tm1c mice with villin-cre mice, in which the expression of the cre recombinase is under the control of the villin promoter. No differences in phosphate homeostasis were found between wild-types and homozygous littermates (Pastor-Arroyo et al., 2020, PubMed ID: 32564464).

Heterozygous:
Heterozygous mice were not analysed, but they are not expected to have any phenotype since homozygous mice did not show any.
