Genetic information | Tetracycline-controlled transgene for the SHANK2A overexpression. The transgene contains a bidirectional tTA-dependent expression cassette which encodes on one site the nuclear localized beta-galactosidase and on the other side the Venus (green fluorescent protein variant) reporter-T2A ribosomal skipping sequence-SHANK2A fusion. The sequence origin for SHANK2A is human. |
Phenotypic information | Homozygous:unknownHeterozygous:No phenotype in absence of the transcriptional activator TA. Upon crossing with TgCam2a-tTA mice, the TgSHANK2A/tTA double-transgenic animals develop an autism spectrum disorder-like phenotype, have a reduced body weight and about 25% show a premature death. |
Breeding history | The line was generated in November 2013 and intercrossed routinely with TgCam2A-tTA mice (B6;CBA-Tg(Camk2a-tTA)1Mmay/J; Stock No: 003010). Double transgenic mice were never used for matings. Heterozygous TgSHANK2A mice were used together with C57BL/6 mice to keep the line. |
