Genetic informationTargeted mice were generated by homologous recombination-based genome editing into embryonic stem cells (ESC). A targeting construct was designed to insert a mCherry red fluorescent protein sequence and a loxP-flanked neomycin (neo) resistance sequence-STOP cassette in the 3' UTR of the X-linked methyl CpG binding protein 2 (Mecp2) gene. The endogenous STOP codon was replaced by a 22bp-linker sequence (Lys-Lys-Asp-Pro-Pro-Val-Ala-Thr). This construct was electroporated into 129P2/OlaHsd-derived E14TG2a ESCs. Correctly targeted ESCs were injected into recipient blastocysts and chimeric mice were bred with C57BL/6 inbred mice. To remove the floxed-neo-STOP cassette female founders were crossed to CMV-cre deleter mice. Mice lacking both the floxed sequence and the cre recombinase transgene were subsequently backcrossed to C57BL/6 for at least 4 generations prior to molecular and phenotypic analysis. Homozygous females and hemizygous males are viable, fertile, normal in size, and do not display any gross physical or behavioral abnormalities. The targeted allele expresses a chimeric protein with the mCherry peptide linked to the C-terminus of both splicing isoforms of the endogenous Mecp2.

Weight gain is less than wild-type littermates between the 5th and 6th week of postnatal life in hemizygous mice (ANOVA test), on a quite pure C57BL/6J background (mutant mice backcrossed for 4 generations to a C57BL/6J inbred strain).
Breeding historyBackcrossed for 4 generations to a C57BL/6 inbred strain. When maintaining a live colony, homozygous females may be bred to hemizygous males. The targeted Mecp2 gene is X-linked.
