Genetic informationGene encoding a protein expressing the human OX40 extracellular domain with mouse intracellular domain knocked into the mouse OX40 genomic locus. The coding sequences of OX40 exon 1 plus some immediate intronic sequences were replaced with a KI cassette, thus the expression of the KI cassette is under control of the mouse Tnfrsf4 gene regulatory element. To engineer the targeting vector, homology arms were generated by PCR using BAC clone RP23-139J18 from the C57BL/6J library as template. In the targeting vector, the Neo cassette was flanked by LoxP sites.
Phenotypic informationHomozygous:
No deleterious changes. They express a receptor with human OX40 extracellular domain, that can be bound by anti-human OX40 monoclonal antibody.

Heterozygous:
Heterozygous mice express half as much hOX40 receptor as the homozygous mice. It also deletes one mouse allele and so expresses 50% of the mouse OX40 receptor.
Breeding historyMice were generated in C57BL/6 background (ES cells).
