Genetic informationIl1atm1a(EUCOMM)Wtsi embryonic stem cells were purchased from the European Mouse Mutant Cell Repository (EuMMCR). Cells from clone EPD0822-4-H02 were cultured in KO-DMEM + KOSR medium + 2i (MEK inhibitor and GSK3 inhibitor) on a gelatin-coated cell culture dish maintained under standard culture conditions (37°C, 5% CO2, humidified). Cells were then microinjected into 4–8 cell B6N-Tyrc–Brd/BrdCrCrl embryos. Surviving embryos were surgically implanted into the oviduct of day 0.5 postcoitum pseudopregnant mice. Resulting black/white C57BL/6N chimeras were backcrossed onto C57BL/6N wild-type mice to assess germline penetrance. Potential founder mice were screened by PCR for LacZ, Neo, and LoxP sites. This line was further crossed with C57BL/6N-Tg(CAG-Flpo)1Afst/Mmucd mice. The FLP recombinase expression provided by this line resulted in a “conditional ready” (floxed) Il1atm1c(EUCOMM)Wtsi allele, in which exon 4 is flanked by loxP sites.
