Genetic informationA 17 kb deletion of the ubinuclein 2 (Ubn2) gene was introduced using CRISPR/Cas9 pronuclear microinjection into zygotes. The deletion spans exons 3-7. CRISPR technology details: the deletion was introduced by cutting the Ubn2 expression vector with two Cas9 RNPs.
Phenotypic informationHomozygous:
We have observed that homozygous Ubn2 mutant males are healthy and fertile, though they are born at a submendelian ratio. In females, homozygosity is mostly embryonic lethal before day 12.5 in the C57BL/6 genetic background, though there is some variability in the phenotype. Rarely, homozygous mutant female are born, but they are infertile. The Ubn1 and Ubn2 mutations contribute additively, whereby the combined mutation of Ubn1 (EMMA strain EM:15975) and Ubn2 (this strain) approximates the Hira (histone cell cycle regulator) lethality which could be expected from the molecular function of both genes.

Heterozygous males and females are healthy and fertile.
Breeding historyMutation was introduced into C57BL/6J background strain. The mutation is maintained in heterozygous state.
