| Breeding history | Hyperovulated C57BL/6J-enriched females were mated with C57BL/6J-enriched males to generate E0.5 embryos. Embryos were harvested from the females and electroporated with Cas9/sgRNA ribonucleoprotein complexes plus a single-stranded DNA template carrying the D260N mutation; 24 hours post-electroporation, embryos were transferred to pseudopregnant recipient females. Founder mice were screened by PCR and Sanger sequencing to identify D260N carriers, and all subsequent breeding was performed in a C57BL/6J-enriched background. |
