Genetic informationTmem38a targeted knock-in carrying the D260N (p.Asp260Asn) missense mutation. TMEM38A-D260N variant has been reported in Emery-Dreifuss muscular dystrophy patients (Meinke P. et al., EBioMedicine, 2020, 51:102587).
Phenotypic informationHomozygous:
Homozygous mice are born at Mendelian rates. No survival, molecular or histological analysis has been carried out.

Heterozygous:
No survival, molecular or histological analysis has been carried out with heterozygous mice.
Breeding historyHyperovulated C57BL/6J-enriched females were mated with C57BL/6J-enriched males to generate E0.5 embryos. Embryos were harvested from the females and electroporated with Cas9/sgRNA ribonucleoprotein complexes plus a single-stranded DNA template carrying the D260N mutation; 24 hours post-electroporation, embryos were transferred to pseudopregnant recipient females. Founder mice were screened by PCR and Sanger sequencing to identify D260N carriers, and all subsequent breeding was performed in a C57BL/6J-enriched background.
