Genetic informationIn the first exon of the Cpa3 gene, the 28 nucleotides coding for the first 9 amino acids were replaced with a codon-improved cre recombinase expression cassette and a Frt-flanked neomycin selection cassette. The cre recombinase expression cassette consists of a splice donor, intron, splice acceptor from rabbit beta-globin and the codon-improved cre sequence (Shimshek et al., Genesis. 2002; doi:10.1002/gene.10023), followed by the rabbit beta-globin polyadenylation site. Transient expression of flp recombinase in a clone of correctly targeted ES cells removed the selection cassette. br bPlease note:/b According to the information provided by the depositor and the submitted material, this strain shares the same targeted allele as the related EMMA strain (EM:16832) but differs in genetic background. The strains can be visually distinguished by their coat colour. Where the distinction between the strains is relevant for experimental use, recipients are advised to confirm the genetic background using appropriate strain-specific genetic markers (e.g. SNP analysis).
Phenotypic informationHomozygous:
No additional phenotype known compared to heterozygous mice.

Heterozygous:
Heterozygous mice are specifically mast cell deficient. Mucosal and connective tissue mast cells are absent. Reduction of basophils. The cre recombinase is expressed in progenitors of the mast cell and basophil lineage (MCBP) and in thymic progenitors (ETP and DN2).
Breeding historyThe original chimera was first crossed to C57BL/6J for germline transmission and a second generation. The colony was subsequently split into two branches: one branch was continuously backcrossed to C57BL/6J to generate the B6-Cpa3tm3(icre)Hrr line (EMMA strain EM:16832), whereas the other branch was established by using an F2 mouse to initiate backcrossing to BALB/c and to generate the C-Cpa3tm3(icre)Hrr line (this strain, EM:16833). The strain has been backcrossed to BALB/c for more than 20 generations. The strain is maintained by HET × WT brother–sister matings.
