Genetic informationWe generated a knock-in mouse line termed LSL-R26Tva-lacZ with concomitant conditional expression of the tumor virus A (TVA) receptor and nuclear lacZ, by targeting the Gt(ROSA)26Sor locus. A loxP flanked transcriptional stop cassette (LSL) was placed 5' of a TVA IRES LacZ-nls expression cassette. The LSL-TVA-IRES-lacZ cassette was inserted into the first intron of the Gt(ROSA)26Sor locus by homologous recombination in ES cells; cre-mediated recombination allows time- and site-specific expression of the TVA receptor for retroviral gene transfer using the RCAS system (replication-competent sarcoma-leukosis virus long terminal repeat with splice acceptor) and the lacZ reporter gene. To demonstrate the ability of this system for conditional somatic gene transfer in vivo, we directed TVA expression to the pancreas. Introduction of an RCASBP(A) vector (Bryan-RSV polymerase and subgroup A envelope viruses) carrying oncogenic KrasG12D induced focal ductal pancreatic lesions that recapitulate human pancreatic intraepithelial neoplasias, which progress to pancreatic ductal adenocarcinomas.
