Genetic informationTargeting construct is derived from 129/Sv DNA. Exon 22 and surrounding intron sequences were replaced by the neomycin resistance gene.
Phenotypic informationKnock-out animals are viable and fertile, although they have an absolute deficit of retinaldehyde oxidase activity in the Harderian gland, decreased retinoic acid levels, and down-regulation of retinoid-dependent genes. Moreover they show thickening of the epidermis in basal conditions and after UV light exposure.
Breeding historyThe knock-out line was obtained upon ES cell injection in B6D2F1 blastocyst and successively backcrossed to C57BL/6N for 9 generations. Heterozygous animals were maintained on C57BL/6N background in SPF facility.
