Genetic informationHomozygous for knock-out mutation (a knock-in of a farnesylated form of EGFP inserted in frame with the two first amino acids of the gene locus). Half of the exon 1 and the entire exon 2 were eliminated and transcription following the EGFP is terminated by a strong polyadenylation signal. A loxP flanked neomycin cassette was included downstream of EGFP for targeting purposes and was subsequently deleted by crossing to a PGK-Cre mouse.
Phenotypic informationThe CD8+ve family of dendritic cells lack expression of DNGR-1 (CLEC9A) and are impaired in the cross-priming of T-cells.
Breeding historyKnock-out generated in the (129S6 x C57BL/6)F1 derived ES cell line S6B6 (equivalent to B6 N1). After crossing 2x to C57BL/6J (Charles River), they were crossed 1x to C57BL/6J PGK-Cre to delete the NeoR gene. The strain is now Cre negative after being crossed a further 7x to C57BL/6J 7x to C57BL/6J Charles River.
