Information from provider

ProviderRamesh PILLAI
Provider affiliationEMBL Grenoble Outstation, European Molecular Biology Laboratory (EMBL)
Genetic informationSingle amino acid point mutation of the ATPase motif (DEAD to DQAD). Presence of loxP sites allows generation of conditional knock-out line.
Phenotypic informationHomozygous:
Heterozygous knock-out mice are already shown to display male-specific infertility (Tanaka SS, et al., The mouse homolog of Drosophila Vasa is required for the development of male germ cells. Genes Dev. 2000 Apr 1;14(7):841-53). We expect our catalytic-dead point mutation of the RNA helicase domain to display similar phenotype: males are infertile in homozygous background, while females of all backgrounds and male of heterozygous background are fertile. I will let EMMA know of the phenotype as soon as I have it in a few months.

Heterozygous:
Heterozygous knock-out mice are already shown to display male-specific infertility (Tanaka SS, et al., The mouse homolog of Drosophila Vasa is required for the development of male germ cells. Genes Dev. 2000 Apr 1;14(7):841-53). We expect our catalytic-dead point mutation of the RNA helicase domain to display similar phenotype: males are infertile in homozygous background, while females of all backgrounds and male of heterozygous background are fertile. I will let EMMA know of the phenotype as soon as I have it in a few months.
Breeding historyFounder mice were crossed with wildtype C57BL/6 females. F1 generation animals are being offered for archiving to EMMA.
