Genetic informationTransgenic mice for the mi-5' gene; 10 copies of the transgene are integrated. The mi-5' construct is a miRNA-expressing gene. The endogenous sequence of the human miR30a gene has been replaced by the targeted sequence (5'-GCCGACGATCTCTTTGACTTCA-3'). The targeted sequence spans over the 5' part of the intermediate early (IE) gene of the Pseudorabies virus (PRV), which currently infects farm animals (especially pigs) and mice. The mi gene is transcribed under the activity of the human eF1alpha gene promoter (-1430, +2). The 5'-HS4 fragment from the chicken beta-globin gene is added as insulator. The construct encompasses also one intron (rabbit beta globin gene, second intron) and several enhancers.
Phenotypic informationExpression of the miRNA targeting the IE gene. Resistance to a Bartha strain PRV infection. No significant innate immune stimulation in non infected animals. Description of the strain is under review.
