Genetic information | The YZ17 line (two copy transgene) contains an additional 130 kb genomic region from mouse distal chromosome 7 spanning from Igf2 to ~40 kb downstream of H19. Igf2 has been inactivated by insertional mutagenesis resulting in an Igf2-LacZ gene fusion. A 1kb conserved intergenic region is flanked with LoxP sites. LacZ activity shows spatial expression of Igf2 in a subset of tissues. The imprinted H19 gene is only expressed when transmitted through the maternal germline. After paternal transmission the gene is repressed. The Igf2-LacZ gene shows appropriate reciprocal imprinting (paternal allele specific expression, maternal repression). Deletion of the intergenic region with Cre recombinase results in partial (mesoderm specific) reactivation of Igf2-LacZ expression following maternal transmission. |
Phenotypic information | The line has not been kept homozygous due to reduced fertility. |
Breeding history | Brother sister matings with sporadic backcrossing to C57BL/6. |
