Genetic information | The AIDel allele has a 28kb deletion on chromosome 17 between 3kb downstream of the Airn transcription start site (TSS) (Igf2r intron 2) and 2.8kb upstream of the Igf2r TSS (GRCm38/mm10, chr17: 12,744,358 -12,772,513). Additionally, Airn is truncated by a polyadenylation cassette immediately before the AIDel deletion. This allele was made by chromosome engineering by targeted meiotic recombination (Herault et al 1998), by breeding together the Airn truncation allele (FVB.129P2-AirnT, Sleutels et al 2002 Nature) and the Igf2r promoter deletion (FVB.129P2-IPD, Sleutels et al 2003 EMBO J.) with Hprt-Cre (B6.129S1-Hprt-Cre, Tang et al 2002 Genesis), and screening the offspring of triple mutant males for recombination events. The AIDel allele should have a 129P2 background near the deletion, and a B6/FVB mixed background in other regions of the genome. The background should be closer to FVB as the mice have been backcrossed to FVB/N for 3 generations. |
Phenotypic information | Homozygous:Homozygous mice should be embryonic lethal between E13.5 and E18.5 due to deletion of the Igf2r promoter (Barlow et al 1991 Nature).Heterozygous:Heterozygotes with a maternally inherited AIDel allele should be embryonic lethal between E13.5 and E18.5 due to deletion of the Igf2r promoter, while mice with a paternal inheritance of the duplication are viable and fertile. |
Breeding history | AIDel mice were derived over 2 generations by breeding together FVB.129P2-AirnT, FVB.129P2-IPD and B6.129S1-Hprt-Cre mice, and then screening the offspring of triple mutant males for recombination events, as described in the “Genetic Description” section. The AIDel founder was then backcrossed to FVB/N for 3 generations. |
