Genetic informationA loxP site was inserted 3' of exon v6 and a neomycin resistance cassette, with a 5' loxP site, was inserted 5' of exon v6 of the Cd44 gene. During insertion, 1.6 kb of sequence was lost in the exon v7 region leading to deficient expression of this variant in homozygous mutant mice (Cd44v7 -/-). The neomycin cassette from pMC1neo was inserted into the EagI site in exon 6 of the Spp1 (secreted phosphoprotein 1; formerly: Opn, osteopontin) gene (a 4.8 kb fragment from 129 mice), that had been cloned into pBluescript (Rittling et al., 1998).
